Research Interests and Accomplishments

Damage to hair cells of our inner ears is a leading cause of deafness and other hearing impairments. Professor Barbara Canlon has spent her career exploring the fundamental workings of the hair cells of the inner ear with the goal of understanding why they are so vulnerable to damage, and ultimately, how they may be protected, repaired, or even replaced.

Pharmacotherapy for inner ear disorders is hampered by the difficulties in local application. Together with researchers at the Swedish Medical Nanoscience Center and Linköping University, a novel technology using organic bio-electronics has been developed to create an interface between electronics and the inner ear. Organic conducting polymers were used as programmable delivery electrodes, in which an electronic in-put signal translates into release of bio-signals (ions, neurotransmitters). Using the peripheral auditory system, we show that the device can selectively stimulate nerve cells responding to glutamate. This technology has great potential as a therapeutic platform, where malfunctioning signal transduction pathways can be modulated by programmable, dose-controlled delivery of signal substances from an implanted bio-electronic device. This new method of communication between electronics and the inner ear will help accelerate the development of therapeutic strategies for hearing disorders. These studies are published in Nature Materials (Simon et al., 2009).

The focus of our resarch is to use luminescent conjugated oligothiophenes (LCOs) to study disease related biological events, such as aggregation of proteins. We are also interesting in using the LCOs as templates for synthesizing novel pharmacopores that can be used as treatment for Alzheimer`s disease and for other protein aggregation disorders.

Local bacterial infections trigger a complex host response whose primary function is to clear the infection and prevent systemic spread of the pathogen. The roles of inflammation and vascular coagulation in this response, and the cross-talk between them is fast gaining recognition as a crucial element in the progression of infectious diseases. How these different systems interact to isolate and clear infection, or the mechanisms by which bacteria disseminate and cause sepsis, are currently unknown. In our work we aim to understand how the innate immune and coagulation systems respond to local bacterial infections during the first hours of infection. We study the first line of these dynamic host responses and how they interact to contribute to infection outcome.

Innovative in vivo infection models combined with intravital imaging are used to visualize infection progression in vivo with single cell resolution. Techniques including transgenic animals, antibody depletion, adoptive transfer, 3D cell tracking and novel imaging probes are used to study the molecular details of the infections.

Our focus currently is on two local bacterial infections that are commonly implicated in sepsis; Staphylococcus aureus skin infection and uropathogenic Escherichia coli kidney infection.

Research Interests and Accomplishments

Our research is oriented in a highly interdisciplinary fashion that has two main directions at The Swedish Medical Nanoscience Center: Theranostic nanoscale polymer based systems for combined imaging and drug delivery , including micelles, crosslinked micelles, dendrimers, and polymer prodrugs mainly focused on breast cancer applications. The second direction is more oriented towards materials science questions in the broader bio and biomedical area, and includes antifouling surfaces and interfaces, resistance against protein deposition etc. The cross-disciplinary research is conducted with partners from both Karolinska Institutet, Royal Institute of Technology and international collaborators.

Background on nanomaterials for drug delivery applications

Functional polymers are an important class of materials that is gaining evermore interest in a wide range of areas extending from nanotechnology, pharmaceuticals, and biomedical applications. Functional polymer assemblies in the size range of 5-100 nm has been identified as key platforms for the development of nanomedical constructs. One of the prime rationales for using nanoparticles and micelle assemblies in biomedical applications is that their 3-D shape presents a discrete and controllable surface to the external environment and that to this surface a variety of bio-active groups capable of interacting with biological systems can be attached. The key challenge is to develop methods which allow for the incorporation of surface accessible and bioavailable ligands, while also providing domains in which therapeutic components can be incorporated within the nanoscale platform. Such constructs aim to deliver the active component tissue selectively to malignant cells and thereby increase therapeutic effects, minimizing toxic responses in healthy tissue and for the patient in general.

Research Interests and Accomplishments

Agneta Richter-Dahlfors (ARD) has a track record in microbial pathogenesis. She was first to visualize the progression of bacterial infection within an organ using confocal microscopy of Salmonella-infected mouse livers. When recruited to Karolinska Institutet in 1998, she focused her research on urinary tract infections. She was first to demonstrate the ability of bacteria to modulate host cell gene expression pattern via bacteria-induced Ca2+ oscillations, and she published a series of papers demonstrating the molecular mechanisms of organ-specific pathogen recognition via epithelial Toll-like receptors in the urinary tract.

By combining advanced surgery and 2-photon microscopy, ARD has pioneered the area of intravital imaging of bacterial infection. These studies emphasize the new concept of “Tissue microbiology”, illustrating bacterial adaptation to the rapidly changing microenvironment as the host initiates its innate defense mechanisms. Inspired by the dynamic in vivo environment, ARD initiated a line of research aiming to develop tools for improved in vitro studies, better mimicking the in vivo situation. Using conducting polymers, she develops systems that translate the languages used in modern electronics (electron as charge carrier) to that of biology (ions, neurotransmitters, peptides etc). ARD published a seminal review on “Organic Bioelectronics” in 2007 demonstrating the use of conducting polymer devices for high-precision biology experiments.

She also demonstrated the usability of such devices in neuroscience applications, and laid the foundation for a new concept, the “artificial nerve cell”. Recently, she demonstrated the first ion transistor, which will become an important device in her attempt to generate chemical circuits for precise cell signaling.
